   Main goal

 

    The same type of tumor can have completely different outcomes in different patients and, alternatively, persons that have been exposed to the same carcinogenic insults (for example, tobacco smoke) during their lives can develop tumors or remain healthy for reasons that remain largely unknown. Our group is interested in understanding the bases of these differences; therefore, the focus of our work is the identification of the genetic and molecular components that determine the different susceptibility, development, response to the treatment and evolution of cancer, among different patients who seemingly have the same hystopathologic disease.

 

    Line of research description


    Cancer is a problem of public health of increasing importance, and we need a better knowledge of the mechanisms that determine the susceptibility, development and evolution of the disease. These depend on the interaction of the genome of the individual with the environment. This interaction determines the variability among patients in the predisposition and development of cancer, as well as in their response to the treatment and evolution. The genetic component that contributes to this variability is constituted by the sum of actions of low-penetrance genes, whose allelic forms interact among them and with the environment to determine the clinical variability among individuals. These genes are named modifier genes and mainly present a pattern of quantitative heredity.


    On the other hand, cancer is not a cellular autonomous process, but it is an aberrant tissue that grows in a not controlled way in the context of the physiology and pathophysiology of a complex organism. And, therefore, it is a disease that not only depends on the properties of the tumour cells, but also of other compartments of the organism, like the immune and endocrine systems, stroma, angiogenesis, etc., which are key pieces in the development and evolution of cancer. Consequently, tumour susceptibility, development and evolution are not only going to be determined by factors intrinsic to the tumour cell involved in processes like proliferation, apoptosis, etc; but they are also going to be influenced by extrinsic factors. Furthermore, these two compartments are not independent and they interact between them, so that the intrinsic factors are capable of recruiting the extrinsic ones, and the availability of the extrinsic factors determines the intrinsic cellular activity. The modifier genes control molecular and cellular factors of these two compartments, and this explains differences in the susceptibility, development and the different clinical evolution between patients who seemingly suffer the same disease.


    Our global aim is to understand the variability in the tumour susceptibility, development and evolution in the global context of the physiology and pathophysiology of the organism, integrating factors both intrinsic and extrinsic to the tumour cell in the same scenario, in order to obtain a global vision of the tumour process at the level of the whole organism. To carry out our goals we use the common technologies of molecular and cellular biology and genetics, together with state-of the-art genomic (expression and CGH arrays) and bioinformatic tools, applied to in vitro and in vivo models (genetically modified mice) and human samples.


    All these technologies are applied in our group to the study of different tumour models, principally of breast, lung and skin cancers and lymphoma, among others. Our final goal is to obtain a better understanding of the molecular and cellular biology of cancer, and the differences among individuals in tumour susceptibility, development and evolution, which will finally result in the development of more individualized clinical applications for the benefit of the patients.
